The Plasmodium parasite is what causes malaria. Around 1891, a search for malaria parasites in birds, reptiles, and mammals was greatly assisted by a new staining method called methylene blue discovered by Yuri Romanovsky in St. Petersburg, Russia. It was used with eosin, fluorescent acidic compounds that bind to and form salts. Researchers used this staining method to examine the blood of malaria patients and noted that the parasites were damaged in patients treated with quinine. Romanovsky concluded that the therapeutic effect of quinine was due to its action on the parasites and not directly on the patient- the first evidence of a drug treatment destroying an infecting microorganism.
